At the neuromuscular junction, stimulations at 15Hz intermittently fail to evoke excitatory junction potentials (EJPs) in almost half of Ube3aUAS.Tag:FLAG third instar larvae with over-expression driven by Scer\GAL4elav-C155, significantly more failures than in controls. There are no significant differences in EJP amplitude over the course of 30 stimulations at 15Hz in larvae that do not exhibit failures. There is no significant difference between controls, Ube3aUAS.Tag:FLAG in mEJP amplitude or frequency in the presence of tetrodotoxin (TTX). In the absence of TTX, there is a significant correlation between amplitude of spontaneous depolarizations seen during mEJP recordings and evoked EJPs in the same larva; the percentage of larvae with spontaneous depolarizations is significantly higher than controls in larvae with Ube3aUAS.Tag:FLAG driven by Scer\GAL4elav-C155.

There is no significant difference in active zone density, bouton numbers or synaptic vesicle number at the neuromuscular junction of third instar larvae with Ube3aUAS.Tag:FLAG driven by Scer\GAL4elav-C155, compared to wild type. Larvae with Ube3aUAS.Tag:FLAG driven by Scer\GAL4elav-C155 have significantly smaller vesicles than wild type. There are also significantly fewer active zones at the neuromuscular junctions of third instar larvae with Ube3aUAS.Tag:FLAG driven by Scer\GAL4elav-C155 (though these numbers could be artificially inflated).

Flies with Ube3aUAS.Tag:FLAG driven by Scer\GAL4GMR.PU have an intermediate rough eye phenotype.

Overexpression of Ube3aScer\UAS.T:Zzzz\FLAG using Scer\GAL4GMR.PF results in a rough eye phenotype.

Expression of Ube3aScer\UAS.T:Zzzz\FLAG in the nervous system under the control of Scer\GAL4elav-C155 results in an increase in dopamine and a concurrent increase in locomotor activity.

A Pur1, PuEY04414, or PuEY02616a heterozygous background enhances the Scer\GAL4GMR.PF-> Ube3aScer\UAS.T:Zzzz\FLAG rough eye phenotype, causing a glazed appearance, loss of inter-ommatidial bristles and a yellowish discoloration, indicative of underlying neurodegeneration.
