Moderate reduction of hbs through the expression of hbsIR.Scer\UAS under the control of Scer\GAL4GMR.PF results in defects in inter-ommatidial cells and bristle group positioning.

Strong reduction of hbsIR.Scer\UAS under the control of Scer\GAL4GMR.PF (in the presence of Dcr-2Scer\UAS.cDa to increase RNAi efficiency) leads to defects in the organization of cone cells and primary cells as well as in inter-ommatidial cells. Most inter-ommatidial cells sort into a single file and ommatidial spacing is not significantly affected. Less than 4% of ommatidia are found in direct contact with each other.

In the developing eyes of hbsIR.Scer\UAS; Scer\GAL4GMR.PF animals at 24 hours APF, ommatidial cores are largely normal, but inter-ommatidial precursor cells (IPCs) fail to sort into a single line as in wild-type. These IPCs have an abnormally relaxed morphology - failing to maximize their contacts with developing primary pigment cells or to minimize their contacts with other IPCs. In addition, inter-ommatidial bristle precursors are often missing or misplaced. The resulting adults have rough eyes dues to disorganized rows of ommatidia.

Simultaneous disruption of sns and hbs through co-expression of snsdsRNA.Scer\UAS and hbsIR.Scer\UAS leads to the direct contact of 27.2% of ommatidia and complete failure of inter-ommatidial cells to sort into single file. These phenotypes represent a strong enhancement of either knockdown alone.

Reduction of both hbs and kirre expression through kirredsRNA.Scer\UAS and hbsIR.Scer\UAS expression under the control of Scer\GAL4GMR.PF enhances slightly the patterning defects seen in kirre down-regulation. Depletion occasionally leads to inter-ommatidial cells occurring in multiple rows between ommatidia. In each case, ommatidial spacing is nonetheless nearly indistinguishable from single knock-down of hbs alone.
