UASt regulatory sequences drive expression of an inverted repeat.
Under control conditions, the adulthood-only expression of traGD764 under the control of Scer\GAL4esg-NP7397 (together with Gal80[ts], for the temporal regulation of expression) does not significantly affect the mitotic index in the adult midgut of males and females, as compared to controls.
Expression of traGD764 under the control of Scer\GAL4elav.PLu (in combination with a Dicer-2 transgene to enhance RNAi efficiency) results in a significant decrease in virgin female receptivity, as compared to controls.
When wild type female flies are exposed to female flies expressing traGD764 under the control of either Scer\GAL4OK72 or Scer\GAL4Desat1.PB (which induces masculinization of the pheromone profile) they exhibit a modestly reduced lifespan compared with females exposed to wild type females.
Female flies expressing traGD764 under the control of Scer\GAL4elav-C155 (co-expressed with Dcr-2Scer\UAS.cDa) exhibit a total loss of the female-specific post-embryonic Ilp7 neurons in the ventral cluster. Embryonic Ilp7 neurons are unaffected. No changes in the Ilp7 neuronal population are seen when traGD764 is expressed under the control of Scer\GAL4Ilp7.PC.
Expression of traGD764 under the control of Scer\GAL4desat1.PB causes the masculinization of female pheromone profiles. As a result, wild-type males show aggression towards females expressing Scer\GAL4desat1.PB-driven traGD764. In contrast, wild-type males do not attack wild-type females.
Females expressing traGD764 under the control of Scer\GAL4elav-C155, unlike wild-type females, show aggression towards bot other females and males. A substantial fraction of wild-type males show aggression towards the females expressing traGD764via Scer\GAL4elav-C155, a behaviour which is not observed towards wild-type females. The pheromone profile of females is unaffected by expression of traGD764 via Scer\GAL4elav-C155. Wild-type males court these females vigorously before transitioning to aggression.
Females expressing traGD764 under the control of both Scer\GAL4desat1.PB and Scer\GAL4elav-C155 exhibit aggressive behaviour towards wild-type males. These females initiate and dominate most fights. Most of the males who lunge at these females do so prior to or without ever copulating. Females expressing traGD764 under the control of both Scer\GAL4desat1.PB and Scer\GAL4elav-C155 show a predominantly male pheromone profile.
Females expressing traGD764 under the control of Scer\GAL41407 show a predominantly male pheromone profile.
Wild-type males show aggression towards females expressing traGD764 under the control of Scer\GAL41407. Copulation is not observed for most of these pairs.
Adults expressing traGD764 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
Expression of traGD764 under the control of Scer\GAL4dsx.KI in XX flies leads to male-like abdominal pigmentation, genitalia and sex combs.
Depending on the insertion line used, expression under the control of Scer\GAL4Mef2.PR can result in viable flies or late pupal lethality.
