The adulthood-only expression of tefuGD11950 under the control of Scer\GAL4esg-NP5130 (in combination with Gal80[ts], for the temporal control of expression) leads to a severe decrease in life-span, as compared to controls; it also results in larger and rounder adult midgut progenitor cells (esg-positive intestinal stem cells and enteroblasts), leads to their disappearance after three weeks of expression, and leads to a significant decrease in the frequency of intestinal stem cells undergoing DNA replication, but does not significantly affect the overall cell density in the adult midgut, the number of intestinal stem cells or the frequency of progenitor cells undergoing DNA replication, as compared to controls. Upon γ-radiation, the adult midgut progenitor cells show a small but evident increase in DNA damage, assessed by pS/TQ and γH2AvD immunofluorescence, as compared to irradiated controls.

When larvae expressing tefuGD11950 under the control of Scer\GAL4ptc-559.1 are irradiated, less cell death is seen in the wing disc in comparison with irradiated controls. The cells in the A compartment are the first to benefit, but the protective effect is able to reach the P compartment.

Adults expressing tefuGD11950 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.

The small and deformed wings in adult flies expressing His1:CG31617NIG.31617R under the control of Scer\GAL4nub.PU are significantly suppressed by co-expression of tefuGD11950.

The adulthood-only co-expression of mei-41KK101792 and tefuGD11950 results in: a more severe increase in γ-radiation-induced DNA damage in adult midgut progenitor cells (esg-positive intestinal stem cells and enteroblasts) compared to either single expression condition; a progressive disappearance of progenitor cells that is comparable to that induced by the expression of mei-41KK101792 alone but more severe than that induced by the expression of tefuGD11950 alone; cell shape changes in intestinal progenitors comparable to those induced by the expression of mei-41KK101792 alone; a decreased cell density in the adult midgut that is comparable to that induced by the expression of mei-41KK101792 alone; decreases in intestinal stem cell number and DNA replication that are comparable to those induced by the expression of mei-41KK101792 alone.
