Animals expressing Unc-89GD14839 under the control of Scer\GAL4Mef2.PR have no regular sarcomeres in the indirect flight muscles at 72hours after pupariation formation (APF), in contrast to what is seen in wild type.
Adults expressing Unc-89GD14839 under the control of either Scer\GAL4Mef2.PR or Scer\GAL4UH3 are flightless, although they show a normal jump response. Myofibrils in the indirect flight muscle are narrower than normal and ultrastructurally, the H-zone is shifted away from the middle of the sarcomere. The thick filaments are asymmetrical in indirect flight muscles expressing Unc-89GD14839 under the control of Scer\GAL4Mef2.PR, with some bare zones far from the middle of the sarcomere. The polarity of the thin filaments follows that of the thick filaments, resulting in abnormally long or short thin filaments. Thin filaments sometimes extend into the H-zone.
Larvae expressing Unc-89GD14839 under the control of Scer\GAL4Mef2.PR show slightly reduced crawling ability compared to wild type, whereas expression under the control of Scer\GAL4UH3 is normal.
Adults expressing Unc-89GD14839 under the control of Scer\GAL4elav.PLu (in the presence of Dcr-2Scer\UAS.cDa to increase the efficiency of RNAi) do not show a significant defect in avoidance of noxious temperature (46[o]C) compared to control flies.
Expression under the control of Scer\GAL4Mef2.PR results in grossly normal adult indirect flight muscles.
