Over-expression of seqScer\UAS.cBa in a wild-type CNS under the control of Scer\GAL4elav-C155; Scer\GAL4sca-109-68 results in the abrogation of ganglionic branching.

The growth cones seen in the axons of R8/R7 photoreceptor cells of pupal stage flies expressing seqScer\UAS.cBa under the control of Scer\GAL4GMR.long fail to undergo the morphological change from an expanded to a condensed appearance. R8 axons extend towards and terminate precisely in the R7 layer (M6), resulting in a single R8/R7 layer in M6 in the medulla of adult flies.

Simultaneous expression of seqScer\UAS.cBa and bnlScer\UAS.cSa in the CNS under the control of Scer\GAL41407 has the same effect as over-expression of bnlScer\UAS.cSa alone.

Expression of seqScer\UAS.cBa under the control of Scer\GAL4GMR.long induces the R7 photoreceptor cells in sev3 mutants to switch from targeting the M3 layer of the medulla to the M6 layer.

CadN405 suppresses the R8 cell targeting defects seen when seqScer\UAS.cBa is expressed under the control of Scer\GAL4GMR.long. The R8 axons no longer terminate in the M6 layer of the medulla and correctly target the M3 layer. However the flies do show an R7 targeting phenotype; the R7 photoreceptors target the M3 layer rather than the M6 layer, as is seen in CadN405 mutants alone.

Comments

Expression of seqScer\UAS.cBa in the ectoderm under the control of Scer\GAL469B rescues the ganglionic branch bifurcation phenotype found in seqC022 and seqZ1241 homozygotes and seqZ1241/seqC022 trans-heterozygotes.

Expression of seqScer\UAS.cBa in midline precursors and glia of seqZ1241, seqC022 and seqZ1241/seqC022 mutants under the control of Scer\GAL4sim.PS accomplishes approximately 50% rescue with those embryos that were not fully rescued showing a considerable attenuation of the phenotype.

Expression of seqScer\UAS.cBa under the control of Scer\GAL4GMR.long rescues the R7/R8 photoreceptor targeting defects seen when sequnspecified mutant somatic clones are induced in R7 cells.
