Randomly generated Mmp2G535R mutant clones rarely populate the tip-cell territory in the air sac primordium.
94% of Mmp2G535R/Mmp2Df animals survive to third instar, all of which pupariate, 47% undergo head eversion, 18% develop head bristles, but none eclose. 74% (n = 23) of Mmp2G535R/Mmp2Df pupae have a melanized lump in the developing thorax, visible as a black spot through the pupal case. The black spot appears before head eversion, varies in size, and remains evident in animals with fully developed legs and bristles.
