Overexpression of timScer\UAS.cYa in PDF neurons under the control of Scer\GAL4P2.4.Pdf does not eliminate circadian rhythms completely, but leads to an increased incidence of arrhythmia. However, no change in fat body triglycerides is observed.
Flies expressing timScer\UAS.cYa under the control of Scer\GAL4tim.PE in a Scer\GAL80Pdf.PS background show rhythmic locomotor activity under constant light conditions, in contrast to wild-type flies, which are uniformly arrhythmic under these conditions.
When timScer\UAS.cYa is driven by Scer\GAL4Mai-60, eclosion rhythms are disrupted. Although peaks are present in the eclosion profile, a rhythm and gating are absent. Adult locomotor activity remains rhythmic as in wild-type. When timScer\UAS.cYa is driven by Scer\GAL4elav-C155 the molecular clock in the lateral neurons is disrupted, and eclosion remains arrhythmic.
When expression is driven in a tim+ background by Scer\GAL4elav-C155, Scer\GAL4Act5C.PI or Scer\GAL4tim.PE arrythmicity occurs. Two copies of P{UAS-tim.Y} give a stronger effect than one. The effect is not evident with Scer\GAL4P2.4.Pdf.

Co-expression of perScer\UAS.cYa and timScer\UAS.cYa under the control of Scer\GAL4P2.4.Pdf results in highly rhythmic flies, with periods close to 24 hours.
The weakened rhythmicity seen in tyfΔG14151 and tyfe00614 flies under constant darkness conditions is not suppressed by expression of timScer\UAS.cYa under the control of Scer\GAL4P2.4.Pdf.

timScer\UAS.cYa driven by Scer\GAL4elav-C155 does not rescue eclosion rhythms, as it does locomotor rhythms in adults.
The degree of rescue of the tim01 locomotor rhythm defects depends to some extent on the site of insertion of the rescuing P{UAS-tim.Y} transgene and is maximally ~70% and minimally 3.1%. One line, P{UAS-tim.Y}3-1 fails to rescue.
