Description

Expression of an aptamer RNA that binds B52 protein is governed by UASt regulatory sequences. The iaRNA gene includes two tandemly repeated pentameric aptameric units. Each unit contains five aptamers which form stem loop structures, followed by a stable tetra loop, followed by a hammer head ribozyme.

Expression of Zzzz\aptamerB52.BBS.Scer\UAS under the control of Scer\GAL4elav.PLu significantly delays larval hatching from embryos and delays the maturation of larval muscle movement: the larvae show an increase in the frequency of uncoordinated muscle twitches that occur at early larval stage (before tracheal filling) before being gradually replaced by coordinated muscle contractions. Expression under the Scer\GAL4eg-Mz360 driver does not impair neuronal cell morphology or number, neither does it interfere with axon guidance or neuronal determination and cell division. Expression of Zzzz\aptamerB52.BBS.Scer\UAS controlled by the Scer\GAL4CY27 driver increases axon extension in embryonic dMP2 neurons.

Flies carrying P{UAS-B52.BBS} show a reduction in viability, even in the absence of Scer\GAL4hs.PB. When expression is governed by Scer\GAL4hs.PB, heat induction causes up to a 50% reduction in survival though surviving flies are morphologically normal. When expression is driven by Scer\GAL4dpp.blk1 salivary glands and wings are normal. When expression is driven by Scer\GAL4G-17 abdominal sternites are normal. When expression is driven by Scer\GAL4I-65 more than 90% of progeny class survive.
