Mutation of the conserved Lys residue within the ATP binding domain to Arg.
air sac tracheoblast of dorsal mesothoracic disc & filopodium
Fewer proliferating air sac tracheoblasts are seen in third instar wing discs of btlKR.hs larvae after heat shock than in wild type controls. Within 3-5 hours of a brief heat shock in btlKR.hs larvae, air sac tracheoblasts of the wing disc exhibit loss of the numerous filopodia that they exhibit in the wild-type. 24 hours after heat shock, these cells produce filopodia as normal.
Heat induced expression in wild type stage 11 embryos causes defects in tracheal primary branching. Induced expression at stage 12 does not affect primary branching but formation of secondary branches is inhibited. Heat induced coexpression of btlKR.hs and btlhs.T:λ\cI-DD at stage 11 almost completely blocks primary branching and coexpression at stage 12 rescues the secondary branching defects.
