In sn28/sn28 nurse cells, the nucleolus has a dispersed phenotype and fewer interconnected tubules than controls. The severity of the phenotype is lower at oogenesis stages 8, 9 and 10A compared to 10B, and in anterior nurse cells.

Macrophage migration is reduced in sn28 mutant stage 15 embryos, with fewer macrophages dispersing from the head to the posterior 2/3 of the embryo along the ventral surface. The macrophages have fewer actin-rich filopodia and microspikes within lamellae.

Nurse cell actin bundle formation is disrupted in sn28 mutants and females flies are sterile.

Stage 14 sn28 embryos show a substantial delay in the migration of plasmatocytes along the ventral midline. 28% of these embryos show "strong" migration defects. These defects persist at later stages - plasmatocytes remain aggregated in the ventral part of the embryo and are unable to reach the posterior region.

The number of plasmatocytes recruited to a laser-induced wound is significantly reduced in sn28 mutants compared to controls. The migratory speed of sn28 plasmatocytes is also reduced.

Plasmatocytes in stage 15 sn28 embryos do not form polarized lamellar extensions, and instead exhibit an abnormal, flacid lamella that surrounds the whole cell. Cell polarity and lamellipodia dynamics are drastically reduced.

Nurse cell nuclei of stage 10 egg chambers are not sustained by actin bundles in sn28 mutants.

The phenotype is suppressed by quaGerm.WT; sn28 females carrying three copies of quaGerm.WT are semi-fertile and produce about 50% the number of eggs produced by wild-type females. Cytoplasmic actin bundles are sparse and disorganised and mature eggs are 60-70% the size of wild-type eggs. sn28 females carrying four copies of quaGerm.WT are fully fertile. The egg phenotype is not rescued by quaGerm.HL.

Bristles severely gnarled.

Expressing snUASp.EGFP under the control of Scer\GAL4VP16.osk rescues the disperse nucleolar phenotype in sn28/sn28 nurse cells.

Expression of snScer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4sn.PZ rescues the macrophage migration defects seen in sn28 mutant stage 15 embryos. The reduction in the number of actin rich filopodia and microspikes within the lamellae of sn28 mutant macrophages is also rescued.

Expression of snScer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 rescues the female sterility and actin bundle formation defects seen in sn28 mutant nurse cells.

Expression of snScer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4sn.PZ rescues the macrophage migration defects seen in Df(1)C128 mutant stage 15 embryos.

Expression of snS289A.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4sn.PZ rescues the macrophage migration defects seen in sn28 mutant stage 15 embryos. The reduction in the number of actin-rich filopodia and microspikes within the lamellae of sn28 mutant macrophages is also rescued.

Expression of snS289A.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 rescues the actin bundle formation defects seen in sn28 mutant nurse cells. Female sterility is restored in 80% of sn28 mutant flies.

Expression of snS289A.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4sn.PZ rescues the macrophage migration defects seen in Df(1)C128 mutant stage 15 embryos.

Expression of snS289D.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4sn.PZ partially rescues the macrophage migration defects seen in sn28 mutant stage 15 embryos.

Expression of snS289D.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4nos.UTR.T:Hsim\VP16 fails to rescue the female sterility and actin bundle formation defects seen in sn28 mutant nurse cells.

Expression of snS289D.Scer\UAS.P\T.T:Avic\GFP-EGFP under the control of Scer\GAL4sn.PZ partially rescues the macrophage migration defects seen in Df(1)C128 mutant stage 15 embryos. The reduction in the number of actin-rich filopodia seen in sn28 mutant macrophages is not rescued.

Scer\GAL4nos.UTR.T:Hsim\VP16-driven expression of snScer\UAS.P\T.T:Avic\GFP-EGFP fully restores the formation of actin bundles around nurse cell nuclei that is normally lacking in sn28 mutants. Female ferility is also restored.

Scer\GAL4nos.UTR.T:Hsim\VP16-driven expression of snS52A.Scer\UAS.P\T.T:Avic\GFP-EGFP fully restores the formation of actin bundles around nurse cell nuclei that is normally lacking in sn28 mutants. Female ferility is also restored.

Scer\GAL4nos.UTR.T:Hsim\VP16-driven expression of snS52E.Scer\UAS.P\T.T:Avic\GFP-EGFP fully restores the formation of actin bundles around nurse cell nuclei that is normally lacking in sn28 mutants. Female ferility is also restored
