Description

14.5kb DNA containing the region -13.5 and -28kb relative to the Ubx transcription start site regulates expression of lacZ.

A 14.5kb fragment of bxd regulatory region of Ubx (coordinates -18.5kb to -4kb on the BX-C walk) is fused upstream of Ecol\lacZUbx.P140.

Presence of trx is able to maintain long term Ubx expression.

Carried in plasmid "Ubx-B", expressed in haploid cell line 1182-4 and the effect of trx and Pc on the Ubx promoter is studied to determine if trx is involved in promoter activation and Pc-dependent repression can be reproduced. Competitive interactions between trx and Pc are also examined in haploid cell line 1182-4 using Ecol\lacZUbx.bxd14.5.
