4kb of 5' flanking DNA, the first (untranslated) exon and the first, large intron up to 350bp upstream of the second exon of D.melanogaster per is fused to approximately 190bp of the putative first intron, 50bp of the non-coding portion of exon 2, and the entire protein coding region, including six introns, of Dpse\per.
Introduction of P{Dpse\perper.P} construct (Petersen, EMBO J. 7: 3939) into per+ individuals successfully rescues circadian arrhythmicity, the circadian locomotor activity profiles resemble those of D.pseudoobscura.pseudoobscura (stronger rhythms with shorter periods).
90% of per01 flies carrying one copy of Dpse\perper.P are arrhythmic, and the reamining 10% have a very weak rhythm. 30% of per01 flies carrying two copies of Dpse\perper.P show clear rescue of rhythmicity, with variable and long free running periods. The activity distribution and rest time values of these flies resemble those of D.pseudoobscura. per+ flies carrying Dpse\perper.P have free running periods the same length as in wild-type D.pseudoobscura.
