Mutant males lack 30-minute courtship memory after 1 hour of conditioning in a courtship conditioning assay. This defect is significantly suppressed if the males are fed 0.1mM 20-hydroxy-ecdysone for 10 minutes immediately prior to conditioning.

The peak dihydropyridine (DHP)-sensitive current in rut2 larval muscles is significantly decreased (78.6 +/- 7.5% of wild-type value) compared to wild type. The current can be increased to 88.2 +/- 3.7% of wild type by application of 1μm forskolin. The peak current is not inhibited by 10 μm H-89 (a PKA inhibitor).

Mutant diminishes cAMP synthesis and reduced the rate of habituation (using the jump-and-flight escape response). Habituation is extremely rapid in dnc rut double mutants.

Reduction in basal level of adult adenylate cyclase. Calcium sensitive. Transheterozygote females dncM14/rut2 and dncM14/rut3 deposit eggs that lack germ cells and display discrete morphological defects.

Homozygous and rut2 dncM14 flies learn about half as well as wild-type controls. Hemizygous flies show essentially wild-type activation and inhibition of adenylate cyclase at various concentrations of calcium ions, although the basal level of adenylate cyclase activity is reduced.

recovered as partial suppressor of the female sterility of homozygous dnc females.

Additional Comments
Genetic Interactions
Statement
Reference

Expression of DopEcRScer\UAS.cIb under the control of Scer\GAL4Mef2.247.Switch in adult mushroom body neurons (using RU486 to limit the timing of expression) restores courtship memory in a courtship conditioning assay in rut2 males.

rut2 partially rescues the egg laying defects of dnc221b/dncM14, but fails to rescue the nurse cell nuclear morphology and 32-cell cyst defects.
