Detailed Description

dnc1/dncML adults show a significant reduction in performance index in an aversive olfactory learning assay compared to control flies.

dncML homozygous and dncM14/dncML heterozygous mutants exhibit an increase in the number of naturally occurring synaptopods at the type II endings. This increase is eliminated in a Dp(1;2)51b background.

Mutant flies show a significant reduction in the number of rest hours per 24 hours compared to controls.

Some dncML flies are arrhythmic with respect to locomotor activity (only 63.16% are rhythmic).

Flies have a low learning score (0.15) in an olfactory classical conditioning paradigm. Homozygous females are sterile. Fertility is restored to approximately 50% if dnchs.PD is expressed using heat shock.

The point of maximal phase delay of the light induced phase response curve (PRC) differs significantly from that of wild type and the circadian period is significantly shorter.

Flies transformed with the P{dnchs.P} or P{rat\dnchs.P} construct display increased learning scores. Reduced learning and memory.

Trans-heterozygotes of Fmr13 and dncML do not exhibit learning deficits.

The low learning score phenotype is completely rescued by Rat\RD1hs.PD expressed using heat shock.

Comments

Expression of dncScer\UAS.cCa exclusively in octopaminergic neurons under the control of Scer\GAL4Tdc2.PC rescues the increase in synaptopods at neuromuscular junction type II endings seen in dncML homozygous and dncM14/dncML heterozygous mutants.

The low learning score phenotype is partially rescued by dnchs.PD expressed using heat shock. Fertility is restored to approximately 50% if dnchs.PD is expressed using heat shock.

A partial rescue is seen after induction of dnchs.P. A significant increase in the initial learning score is seen. Heat shock pulses four days after hatching partly rescues female fertility.
