Centrosome number is abnormal in mutant larval neuroblasts, with one to three centrosomes being seen.
The larval brains of aur87Ac-5 homozygotes, as well as those of aur87Ac-5/Df(3R)T-47, aur87Ac-5/Df(3R)T-61 and aur87Ac-5/Df(3R)P-79 animals, have a significantly elevated mitotic index with the majority of cells delayed in a metaphase-like state. A substantial proportion of metaphase chromosomes seen in these cells are arranged in circular arrays (indicating monopolar spindles). A small proportion of cells in these brains are aneuploid or polyploid. Note, the hemizygotes have less cells with circular arrays than homozygotes, suggesting that aur87Ac-5 might be neomorphic. In dividing aur87Ac-5/Df(3R)T-61 larval brain neuroblasts, astral microtubules are short and reduced in number throughout mitosis compared to wild-type. Many of the spindles in these cells are bipolar but mono-astral, with the remaining diminutive aster nucleated around a fragmented centrosome, frequently only present at one pole. Poles of bipolar spindles in such mutant brains either showed focused microtubules with no evidence of an organized centrosome, or spindle microtubules nucleated from bodies containing multiple centrioles.
aur87Ac-5/aur87Ac-3 larval brain neuroblasts show very few normal metaphase and anaphase figures, and many circular mitotic figures. Larval brain neuroblasts from aur87Ac-4/aur87Ac-5 show multiple mitotic figures with only one centrosome in the circular mitotic figures. The single centrosome is larger than those of bipolar spindles, consistent with them being an aggregate of duplicated centrosomes.
