tetO regulatory sequences drive expression of the tTA transcriptional activator. In the absence of tetracycline, the tTA protein produced by this transgene binds its own (tetO) regulatory sequences, resulting in positive feedback and high levels of tTA expression. In the presence of dietary tetracycline, the tTA protein cannot bind the tetO sequences, suppressing expression of tTA from the transgene.
