A reporter is used to indicate N signalling. In late third instar wing discs, there is a complex expression pattern including a prominent stripe at the dorsal ventral (DV) wing disc border and a two-cell-wide stripe perpendicular to the DV boundary coincident with the anterior posterior (AP) organizer. The staining is on the ci-expressing side of the border indicating expression on the anterior side of the AP border. As development advances, expression decreases at the dorsal ventral border and increases at the AP border. When the pupal wing has fully everted, a number of stripes of lacZ expression parallel to the AP border are visible; the strongest of these stripes is at the AP border.
EcollacZDdc.be2.E(spl)m8-HLH transcript is observed in peripheral cells, a cell in the larval midgut that extends processes around midgut imaginal islands.
The N signaling reporter is expressed at multiple steps in the follicle stem cell (FSC) lineage. It is expressed in an FSC and a cross-migrating FSC daughter, a cell between the two region 2b cysts, a cell at the posterior of the second region 2b cyst, multiple stage 6 follicle cells, polar cells, and cap cells.
EcollacZDdc.be2.E(spl)m8-HLH protein is observed in 18% of small cells that occupy a basal position within the adult midgut epithelium.
EcollacZDdc.be2.E(spl)m8-HLH protein is observed in half of ScerGAL4esg-NP5130-expressing cells in the adult midgut epithelium.
In third instar larva, high levels of expression in the imaginal discs, in pattern similar to E(spl)mβ; high levels at tracheal branchpoints.
