Description

When fused to the N-terminal end of a protein of interest, Tag:TDegX acts as a conditional N-degron sequence that can be activated by TEVp::p14. Tag:TDegX consists of a TEVp recognition site (ENLYFQ-X) where X is a destabilizing residue (one of F, D, K, H, Y), a N-degron sequence (HKSGAWKLPVSLVK) and a protein interaction domain (residues 381-424) from the Homo sapiens SF3B1 gene (HGNC:10768) - this interaction domain is specifically bound by the Homo sapiens 'p14' gene (SF3B6, HGNC:30096). In this form, the N-degron sequence is dormant as it is protected by the TEVp recognition site. The TEVp::p14 artificial protease can bind to Tag:TDegX (via interaction between the SF3B1 domain in Tag:TDegX and the 'p14' sequence in TEVp::p14), resulting in efficient cleavage at the TEVp recognition site that exposes the destabilizing X residue, which becomes the N-terminal amino acid. This activates the N-degron sequence, resulting in degradation of the tagged protein of interest via the ubiquitin-proteasome system. To monitor the cleavage, a reporter such as a fluorescent protein can be fused to the protein of interest, N-terminal to the Tag:TDegX sequence (PMID:19401679, PMID:22350916).
