A set of transgenic insertion stocks derived by TE mobilization using the P-element construct P{GawB}. The P{GawB} construct carries a w+mW.hs mini-white visible marker and Scer\GAL4 driver/enhancer trap sequences. The GAL4-UAS system is a binary system using Scer\GAL4, a yeast transcription activator protein gene, and Scer\UAS, the DNA binding site for the GAL4 protein. The P{GawB} construct acts as an enhancer trap: expression of the GAL4 is sensitive to enhancers in the genomic region of an insertion. When paired with a construct or insertion carrying a reporter gene downstream of Scer\UAS, the reporter gene is driven by and reflects the expression of the GAL4.
qsm reporter expression is observed in clock neurons. It is most strongly expressed in the 2 DN2, 4-17 DN3, 1-2 LNd, and 1 LNv neurons. Some but not all of the clock neurons that express qsm also express cry. 1-2 LNd and 1-2 of the DN3 neurons, 1 DN1 neuron, and the single fifth s-LNv neuron coexpress cry while 2 DN2 neurons and ~10 DN3 neurons are cry negative. Reporter expression is observed in wide neuronal arborizations covering an area of the antennal lobes, tritocerebrum, and suboesophageal ganglion. Ramifications in the the contralateral antennal lobes are interlinked by many short connectives and a prominent axonal bundle runs to the antennal nerve. Arborizations in the tritocerebrum and the suboesophageal ganglion are connected with nerves innervating the mouthparts and the ventral cord. A number of stained neurons are located along the antennal lobe-tritocerebrum-suboesophageal ganglion arborization. Neurons in the posterior pars intercerebralis and neurons in lateral and ventral parts of the suboesophageal ganglion are labelled. Signal is also seen in a bundle of ramified fibers that interlink the contralateral optic lobes passing through the lateral and dorsal brain. Neurons innervating this link are located in regions containing clock neurons. In the optic lobes, ramified fibers are visible in a middle layer of the medulla.
