I. P{GawB}GII5 is homozygous viable and fertile second chromosome insertion. In combination with a P{UAS-lacZ} insertion*, staining is seen in embryonic peritracheal cells and pericardial cells. (*Note: The specific identities of the P{UAS-lacZ} and P{UAS-Notch} constructs were not provided.) II. P{GawB}G11-1 is a homozygous and hemizygous viable and fertile X chromosome insertion. In combination with a P{UAS-lacZ} insertion, staining is seen in a subset of the embryonic peripheral nervous system and uniformly in stage 9 follicle cells. III. P{GawB}112A is a homozygous and hemizygous viable and fertile X chromosome insertion generated in the lab of Y.N. Jan. In combination with a P{UAS-lacZ} insertion, staining is seen in the embryonic epidermis and in the stalk cells and in follicle cells at the anterior and posterior poles of the egg chamber. IV. P{GawB}109C1 is a homozygous and hemizygous viable and fertile X chromosome insertion generated in the lab of Y.N. Jan. In combination with a P{UAS-lacZ} insertion, staining is seen in the embryonic epidermis and in the follicle cells at the anterior and posterior poles of the egg chamber. V. The following insertions were generated by mobilization of P{GawB}109C1 in the lab of Y.N. Jan. A. P{GawB}109-28 1. A homozygous viable and fertile second chromosome insertion. 2. In combination with a P{UAS-lacZ} insertion, staining is seen in the follicle cells of the dorsal appendage. B. P{GawB}109-30 1. This insertion is carried on the second chromosome. 2. In combination with a P{UAS-lacZ} insertion, staining is seen in the embryonic dorsal midline and peripheral nervous system, the larval ventral nerve cord and segmentally in the nervous system, and in stalk cells, follicle cells at the poles of the egg chamber and the precursors to the follicle cells in the germarium. 3. In combination with P{UAS-N}, it is larval lethal. 4. In combination with P{UAS-Cdc42.L}, it is larval lethal. C. P{GawB}109-39 1. This insertion is carried on the third chromosome. 2. In combination with a P{UAS-lacZ} insertion, staining is seen in blastoderm stripes, and in stage 2 follicle cells. 1. In combination with P{UAS-N}, it is larval lethal. 2. In combination with P{UAS-Cdc42.L}, it is larval lethal. 3. In combination with P{UAS-Rac1.L}, it is larval lethal. D. P{GawB}109-53 1. This insertion is carried on the third chromosome. 2. In combination with a P{UAS-lacZ} insertion, general staining is seen in embryos. In ovaries, staining is seen in stalk cells and terminal filament cells. 1. In combination with P{UAS-numb.W}, it is embryonic-larval lethal. 2. In combination with P{UAS-N}, it is larval lethal. 3. In combination with P{UAS-Cdc42.L}, it is larval lethal. E. P{GawB}sca109-68 (FBti0004036) 1. A homozygous lethal, second chromosome insertion. 2. In combination with a P{UAS-lacZ} insertion, staining is seen in proneural cells of the wing imaginal disk and in stage 6 follicle cells. 3. In combination with P{UAS-numb.W}, it causes bristle defects. 4. In combination with P{UAS-N}, it causes bristle defects. 5. In combination with P{UAS-Cdc42.L}, it is larval lethal. 6. In combination with P{UAS-Rac1.L},it causes bristle defects. F. P{GawB}109-69 1. This insertion is carried on the second chromosome. 2. In combination with a P{UAS-lacZ} insertion, staining is seen in embryonic dorsal epidermis and in parts of the peripheral nervous system, in the larval wing imaginal disk and parts of the brain, and in stage 8 follicle cells. 3. In combination with P{UAS-Cdc42.L}, it is larval lethal. 4. In combination with P{UAS-Rac1.L}, it causes rough eyes. G. P{GawB}109-79 1. A homozygous viable and fertile second chromosome insertion. 2. In combination with a P{UAS-lacZ} insertion, staining is not seen in embryos, but strong staining is seen in stage 2-3 follicle cells. 5. In combination with P{UAS-numb.W}, it is embryonic-larval lethal. 6. In combination with P{UAS-N}, it is larval lethal. 7. In combination with P{UAS-Cdc42.L}, it is larval lethal. 8. In combination with P{UAS-Rac1.L},it is embryonic-larval lethal. VI. The following insertions were generated in the lab of Gerhard Technau. A. P{GawB}1158x 1. A homozygous and hemizygous viable and fertile X chromosome insertion. 2. In combination with a P{UAS-lacZ} insertion, staining is seen in embryonic ventral muscles and a subset of the CNS. B. P{GawB}60IIA 1. A homozygous viable and fertile second chromosome insertion. 2.
