      Description

      A set of transgenic constructs that each consist of single sgRNA that contains MS2 loops. The tagged sgRNA forms one component of the three-component flySAM (synergistic activation mediator) system, which is designed to produce effective transcriptional activation of the gene targeted by the sgRNA. In this collection, each construct also contains the other two components of the flySAM system (nuclease-dead Cas9 fused to a transcriptional activation domain plus an MCP-tagged transcriptional activator), and so target gene expression can be activated by crossing to any GAL4 line.

      Many of the TRiP RNAi and sgRNA stocks generated at Harvard Medical School (HMS lines) and Tsinghua University (HMC lines), but not stocks generated at the National Institute of Genetics in Japan (HMJ lines), contain the sev21 allele as a result of introducing a mutated X chromosome from Bloomington stock 35781 (FBst0035781) or 32261 (FBst0032261) as the transgenes were being established in stock.
