Genomic sequence has been inserted into the attP site in TI{TI}chers24, reconstituting a cher locus containing mutations that result in a protein in which the mechanosensor region (MSR) is shifted to the closed state (the masking β strand at each of the two interaction sites within the MSR has been substituted with the corresponding residues of a high affinity sequence from human platelet glycoprotein Ib, resulting in a sequence that requires higher forces, 6-12 pN, for the unmasking of the interaction sequence compared to the native sequence). The cher coding sequence is tagged at the C-terminal end with mGFP6.
