The RMCE cassette of the Mi{MIC}ChATMI08244 insertion (located in a VAChT intron) has been replaced with a phase 1 Trojan p65AD cassette that encodes a splice acceptor site followed by the T2A peptide, sequence encoding a p65(AD)::Zip+ hemidriver and an Hsp70 transcription termination signal. This results in expression of a truncated VAChT transcript lacking the 3'UTR, together with expression of the p65(AD)::Zip+ hemidriver under the control of endogenous VAChT regulatory sequences.
The attP cassette in the Mi{MIC}ChATMI08244 insertion has been replaced with a Trojan p65AD cassette that contains a splice acceptor site followed by the T2A peptide and the p65(AD)::Zip+ component of the SplitGAL4 system, and an Hsp70 transcription termination signal.
