Description

A small deletion early in the coding sequence that causes a frame-shift and the premature termination of the chk protein without a destabilization of the mRNA - this results in a specific decrease of chk (without an effect in the second ORF (CG45092) of this dicistronic mRNA.

Comment:

A 17 bp deletion shortly after the chk ATG causes a frameshift and early translation termination after the addition of 10 novel amino acids.

chkCRSPR as well as chkCRSPR/Df(2R)BSC326 mutants do not show any obvious defects in the gross adult brain morphology or any deficit in larval locomotion (assessed by speed of crawling and frequency of contraction) but display significant decrease in adult climbing ability. chkCRSPR homozygotes exhibit synaptic transmission defects (increased amplitude of evoked postsynaptic currents at 1 or 2 mM external Ca[2+], no difference in amplitude is observed at low calcium levels) at larval neuromuscular junctions, have reduced adult survival rate and lower resistance to starvation (also observed with chkMB04207/chkCRSPR transheterozygotes) as compared to controls.
