The attP cassette in the Mi{MIC}VGlutMI04979 insertion has been replaced with a Trojan 3XGAL80 cassette that contains a splice acceptor site followed by three copies of Scer\GAL80 (in detail this consists of the T2A peptide - Scer\GAL80 tagged with both T:Zzzz\FLAG and T:Ivir\HA1 - a P2A peptide - Scer\GAL80 tagged with T:Avic\GFPEGFP - a T2A peptide - an untagged copy of Scer\GAL80 - an Hsp70 transcription termination signal). The insertion is in the correct orientation to gene trap VGlut. The splice acceptor ensures that the T2A-GAL80-P2A-GAL80-T2A-GAL80 open reading frame is incorporated into the mRNA of VGlut, while the T2A and P2A sequences promote the separate translation of each Scer\GAL80 open reading frame (the first T2A sequence should also result in the truncation of VGlut). Thus Scer\GAL80 expression is driven by the native VGlut regulatory sequences.
