Description

Insertion of a Mi{Hto-WP} element upstream of zfh2, at coordinate 4:515046 (release 5 genome). The HTO exon in the Mi{Hto-WP} element splices to the zfh2 exon that begins at coordinate 5:536208 . This zfh2 exon consists of 5'UTR, but is in frame with the zfh2 codon in the next exon. This results in a fusion protein being expressed under the control of Scer\UAS regulatory sequences in the Mi{Hto-WP} element. The fusion protein consists of 3 copies of the T:Zzzz\FLAG tag, the mCherry form of Disc\RFP, linker sequence translated from the zfh2 5'UTR exon (KTPSPVRGNYAIDSINWSNNGNDDGSP) and the zfh2 coding sequence.

Expression of zfh2EAB under the control of Scer\GAL4Su(H).GBE (in combination with tub-Gal80[ts]) is sufficient to drive activation and growth of enteroblasts.

Expression of zfh2EAB under the control of Scer\GAL4esg.PU (in combination with tub-Gal80[ts] to restrict expression to the adult stage) leads to an increase in the cell size of enteroblasts in adult guts when compared to controls.

Expression of zfh2EAB under the control of Scer\GAL4Su(H).GBE (in combination with tub-Gal80[ts]) leads to activation of enteroblasts (by decreasing cell circulatory and increasing number of cells with membrane protrusions); an increase in the cell size and cell volume of enteroblasts; and an increase in the proliferation of intestinal stem cells in adult guts when compared to controls.

Expression of zfh2EAB under the control of Scer\GAL4esg.PU (in combination with tub-Gal80[ts]) leads to formation of multilayered intestinal epithelium in the anterior and posterior midgut when compared to controls. Its expression under the control of Scer\GAL4Su(H).GBE (in combination with tub-Gal80[ts]) also leads to formation of multilayered intestinal epithelium in posterior midgut.

Enteroblast clones expressing zfh2EAB (under the control of Scer\GAL4T80) do not exhibit terminal differentiation when compared to controls.

Expression of one copy of zfh2EAB under the control of Scer\GAL4GMR.PU produces flies that have mildly glazed (lacking clear separation of ommatidia) and rough eyes.

Expression of zfh2EAB under the control of Scer\GAL4Bx-MS1096 severely reduces growth of the wing blade.

Expression of zfh2EAB under the control of Scer\GAL4ptc-559.1 prevents the proper extension of the leg, and the tarsal region is not properly patterned or segmented.
