Expressing parkHMS01800 under the control of Scer\GAL4ple.PU leads to adults showing a progressive decrease in climbing ability;

Expressing parkHMS01800 under the control of Scer\GAL4Mef2.PR disrupts the mitochondrial network, with increased mitochondrial aggregates and increased number of mitolysosomes, in both adult and larval muscles, and wing posture defects, and wing posture defects.

Expression under the control of Scer\GAL4αTub84B.Switch.PK (induced by RU486 treatment) results in wing posture defects, decreased adult climbing activity and disorganization of muscle intestine fibers.

Expression under the control of Scer\GAL4elav.PLu leads to a decrease in the number of PPL1 neurons and a decrease in mitolysosomes in the larval and adult brain; there is decreased climbing activity.

The eyes of adults expressing parkHMS01800 under the control of Scer\GAL4GMR.PF show a progressive loss of rhabdomeres (more frequent at day 30 than at day 2).

Expression of parkHMS01800 under the control of Scer\GAL4how-24B results in deficit in larval locomotion: the crawling speed, distance from start to finish as well as the total length travelled over time are all significantly reduced in the knock-down third instar larvae compared to controls.

Expression of parkHMS01800 under the control of Scer\GAL4tin.CΔ4 evokes heart tube hypocontractility one and four weeks after eclosion, and increases cardiomyocyte mitochondrial size. Lifespan is similar to controls. The flies display an accelerated age-related decline in climbing performance.

Expression of SodScer\UAS.cWa suppresses the heart tube hypocontraction and increase in mitochondrial reactive oxygen species (ROS) seen when parkHMS01800 is expressed under the control of Scer\GAL4tin.CΔ4.

Expression of Sod2Scer\UAS.cMa suppresses the heart tube hypocontraction and increase in mitochondrial reactive oxygen species (ROS) seen when parkHMS01800 is expressed under the control of Scer\GAL4tin.CΔ4.

Expression of CG6878KK108566 suppresses the heart tube hypocontraction and increase in mitochondrial reactive oxygen species (ROS) seen when parkHMS01800 is expressed under the control of Scer\GAL4tin.CΔ4.

Co-expression of MarfmiRNA.cUa.Scer\UAS and parkHMS01800 under the control of Scer\GAL4tin.CΔ4 suppresses the heart tube cardiomyopathy and some of the mitochondrial defects seen when either transgene is expressed alone. The proportion of reactive oxygen species (ROS)-producing and depolarized organelles is reduced, and the reduction in mitochondrial DNA content seen in flies expressing parkHMS01800 is rescued. However, the mitochondria show characteristics of both parent lines: they are still enlarged as is seen with parkHMS01800, and appear fragmented as is seen with MarfmiRNA.cUa.Scer\UAS.
