Individuals expressing cazUAS.x10.Tag:FLAG under the control of Scer\GAL4elav-C155 do not show any obvious change in lifespan, as compared to controls. cazKO hemizygous males expressing cazUAS.x10.Tag:FLAG under the control of Scer\GAL4elav-C155, however, exhibit significant decreases in lifespan and climbing ability (i.e. speed), as compared to controls.

The expression of cazScer\UAS.x10.T:Zzzz\FLAG under the control of Scer\GAL4CCAP.PI leads to a decrease in axonal transport of NPY-positive vesicles (i.e. a significant decrease in anterograde transport and a significant increase in the stationary fraction) and of mitochondria (i.e. a significant decrease in retrograde transport and a significant increase in the stationary fraction), in third instar larval motor neurons, as compared to controls.

The expression of cazScer\UAS.x10.T:Zzzz\FLAG under the control of Scer\GAL4Toll-6-D42 does not lead to any obvious defects in viability or third instar larval locomotion, but induces a significant and progressive decrease in adult locomotion, as compared to controls.

Overexpression of cazScer\UAS.x10.T:Zzzz\FLAG in motor neurons via Scer\GAL4RapGAP1-OK6 induces neuromuscular junction (NMJ) expansion.

Overexpression of cazScer\UAS.x10.T:Zzzz\FLAG has no effect on longevity.

Neuronal overexpression of cazScer\UAS.x10.T:Zzzz\FLAG under the control of Scer\GAL4elav-C155 in TBPHΔ23/Df(2R)BSC660 mutants restores their eclosion rate and longevity to levels not significantly different from that of wild-type animals.

Although overexpression of cazScer\UAS.x10.T:Zzzz\FLAG under the control of Scer\GAL4elav-C155 in TBPHΔ23/Df(2R)BSC660 mutants significantly improves their locomotion velocity, these animals are still significantly slower than controls.

The level of NMJ expansion induced by the overexpression of cazScer\UAS.x10.T:Zzzz\FLAG via Scer\GAL4RapGAP1-OK6 is not affected by TBPHΔ23/Df(2R)BSC660.

Comments

Expressing cazUAS.x10.Tag:FLAG under the control of Scer\GAL4elav-C155 rescues both caz2 and caz[KO] hemizygous males' pupal lethality, but the resulting adults exhibit a significant decrease in climbing ability (i.e. speed) as compared to controls.

The expression of cazScer\UAS.x10.T:Zzzz\FLAG under the control of Scer\GAL4CCAP.PI rescues the decreased axonal transport of mitochondria and (NPY-positive) vesicles observed in caz383 homozygous third instar larval motor neurons.

The expression of cazScer\UAS.x10.T:Zzzz\FLAG under the control of Scer\GAL4Toll-6-D42 rescues the semi-lethality, the decreased third instar larval crawling capacity and the decreased adult locomotor capacity observed in caz383 homozygotes.

Expression of cazScer\UAS.x10.T:Zzzz\FLAG under the control of Scer\GAL4elav-C155 partially rescues the lethality seen in caz2 or cazKO hemizygous males.

Expression of cazScer\UAS.x10.T:Zzzz\FLAG in the nervous system under the control of Scer\GAL4elav-C155 rescues the adult viability of males hemizygous for the 'caz1' mutant chromosome (Df(1)383 onto which a transgene containing CG32576+t5.0 has been recombined to provide CG32576[+] function) to 83.7% of control levels. The locomotion defects seen in caz1/Y adult escapers are also strongly, but not completely rescued in these animals, while longevity is completely rescued.
