UAS regulatory sequences drive expression of a short inverted repeat.
testis, with Scer\GAL4C587
The adulthood-only expression of E(z)HMS00066 under the control of Scer\GAL4C587 and Gal80[ts] leads to adult testes exhibiting significant increases in the numbers of cyst stem cells and of early stage germline cells, as compared to controls.
Expression of E(z)HMS00066 under the control of Scer\GAL4nos.PG results in complete sterility in females. Oogenesis is arrested before the vitellogenic stages, resulting in the presence of degenerating stage 6 egg chambers in the ovary. These mutant egg chambers do not contain an oocyte nucleus, instead having 16 nurse-like cells. At stage 4, the mutant oocytes do not have a compact karyosome and rather resemble nurse cells in size and shape.
Cells undergoing ectopic endocycles (ectopic S phases in the absence of mitotic markers) are seen region 2b of the germarium in females expressing E(z)HMS00066 under the control of Scer\GAL4nos.PG.
