    Drives expression in adult mushroom body anterior paired lateral cell (APL), and in the Optix-expressing compartment of the outer optic anlage at larval stages.

    ScerGAL4NP2631 drives expression in the dorsal eye disc in a pattern recapitulating that of Optix. In the adult head, ScerGAL4NP2631 drives expression in a medial stripe running from the anterior edge of the dorsal head capsule to the anterior ocellus. ScerGAL4NP2631 does not drive expression in the embryonic primordium of the eye-antennal disc.

    ScerGAL4NP2631 drives expression that overlaps with Optix. In the adult, it is expressed in much of the cell body rind of medulla and part of the lamina with a clear central gap that corresponds to anterior boundaries seen in larval Optix expression.

    ScerGAL4NP2631 is expressed in a subset of neurons in the adult brain, including the median bundle, protocerebral bridge, subesophageal ganglion and the mushroom body anterior paired lateral cell.

    Optogenetic activation (using Crei\ChR1ReaChR.UAS.FRT.mCherry.Citrine with 590 nm (yellow) light) of the set of neurons expressing Scer\GAL4NP2631 or Scer\GAL4GMR22H05 that also express Scer\FLP1fru in adult males results in the induction of pulse and sine song in the absence of a female.

    Thermogenetic activation (using TrpA1FRT.stop.UAS.Tag:MYC at 31[o]C) of the Scer\FLP1fru-expressing subset of Scer\GAL4NP2631 cells in adult males does not induce wing extensions, compared to controls.

    Thermogenetic activation (using TrpA1-FRT.UAS.cvPa) of neurons targeted by the intersection of the Scer\GAL4nSyb.PS and Scer\FLP1fru expression patterns induces courtship singing in isolated flies at temperatures above 28.5[o]C in males and 31.5[o]C in females. These songs exhibit more widely spaced pulses than those of natural songs, and are often polycyclic in females.

    Thermogenetic activation (using TrpA1-FRT.UAS.cvPa) of neurons targeted by the intersection of the Scer\GAL4NP2631 and Scer\FLP1fru expression patterns results in reliable male courtship song singing at temperatures between 30[o]C and 33[o]C. Males produce monocyclic pulses organized into distinct trains that are indistinguishable from those of natural courtship songs. The interpulse interval of these songs elicited at 30[o]C are comparable to those of natural songs from wild-type flies at the same temperature.

    Thermogenetic activation (using TrpA1-FRT.UAS.cvPa) of neurons targeted by the intersection of the Scer\GAL4VT040556 and Scer\FLP1fru expression patterns results in thermally induced courtship songs consisting of monocyclic pulses organized in trains with interpulse intervals within the natural range.

    Thermogenetic activation (using TrpA1-FRT.UAS.cvPa) of neurons targeted by the intersection of the Scer\GAL4VT041688 and Scer\FLP1fru expression patterns typically results in approximately 50% of males producing pulse songs when warmed above ~31.5[o]C. These songs are organized into distinct trains of monocyclic pulses, but with significantly longer interpulse intervals.

    Thermogenetic activation (using TrpA1-FRT.UAS.cvPa) of neurons targeted by the intersection of the Scer\GAL4VT043702 and Scer\FLP1fru expression patterns elicits wing extension but not wing vibration. Moreover, these wing extensions are often bilateral or, if unilateral, persistently involve one or the other wing.

    Thermogenetic activation (using Ctet\TeTxLCUAS.cSa) of fru neurons (intersection of Scer\FLP1fru and Scer\GAL4NP2631 expression patterns) results in males singing courtship songs less often and with fewer pulse trains. They are also less successful in their courtship attempts.

    Thermogenetic activation (using Ctet\TeTxLCUAS.cSa) of fru neurons (intersection of Scer\FLP1fru and Scer\GAL4VT040556 expression patterns) results in males singing courtship songs less and copulating less than controls.

    Thermogenetic activation (using Ctet\TeTxLCUAS.cSa) of a subset of fru neurons in the VNC: dPR1, dMS7 and a heterogeneous set of cells in the abdominal ganglia (intersection of the Scer\FLP1fru and Scer\GAL4VT041688 expression patterns) results in a significant reduction in both courtship song production and copulation success when males are paired with wild-type virgins. The frequency of wing extension is however similar in both test and control males.

    Thermogenetic activation (using Ctet\TeTxLCUAS.cSa) of neurons targeted by the intersection of the Scer\FLP1fru and either Scer\GAL4VT019579 or Scer\GAL4VT005534 expression patterns produces fewer singing males (when paired with virgin females) and those that do sing do so less than in controls. Test males are also less successful in mating.

    Thermogenetic activation (using Ctet\TeTxLCUAS.cSa) of neurons targeted by the intersection of the Scer\FLP1fru and Scer\GAL4VT043702 expression patterns results in a reduction in pulse song (with fewer than half of them producing any pulse song at all), and wing extension, with most failing to copulate. The songs of these flies exhibit significantly longer interpulse intervals than in wild-type and their pulses are frequently polycyclic.
