Description

UASt regulatory sequences drives expression of a truncated Shaw protein (a stop codon has been introduced at codon 369) which is tagged at the C-terminal end with Tag:FLAG. The protein produced is predicted to be a dominant negative form of Shaw.

Expression of Shawtr.Scer\UAS.T:Zzzz\FLAG in all clock neurons (under the control of Scer\GAL4tim.PE) has no clear effect on locomotor rhythms in LD. The same is true when Scer\GAL80Pdf.PS or Scer\GAL80cry.PS are added to restrict expression.

Expression of the dominant-negative Shawtr.Scer\UAS.T:Zzzz\FLAG in the DNs alone, under the control of Scer\GAL4tim.PE and Scer\GAL80cry.PS does not result in DD period lengthening.

Males overexpressing Shawtr.Scer\UAS.T:Zzzz\FLAG with Scer\GAL4C164 or Scer\GAL4elav-C155 are less viable than female siblings. Some of these adults show an ether-induced shaking phenotype. Some adults show a phenotype with unfurled wings and softened or abnormally tanned cuticle. Some small pupae fail to eclose. These phenotypes are also obtained with high penetrance when driving Shawtr.Scer\UAS.T:Zzzz\FLAG with Scer\GAL4Ccap.PP.

Flies overexpressing Shawtr.Scer\UAS.T:Zzzz\FLAG with Scer\GAL4Mhc.PW show an ether-induced shaking phenotype.

Females expressing Shawtr.Scer\UAS.T:Zzzz\FLAG with Scer\GAL4elav-C155 show a reduced body weight compared to controls.

Expression of Shawtr.Scer\UAS.T:Zzzz\FLAG with Scer\GAL4C380 depolarises resting membrane potential and increases input resistance of a larval motor neuron (MNISN-Is), decreases the spike rate and increases the delay to first spike. Injection of current to hyperpolarize these neurons to -60mv leads to a significant normalisation of their firing properties, but not to wildtype levels.
