UAS regulatory sequences drive expression of SuUR amino acid residues 495-962, which are tagged at the N-terminal end with Tag:HA and with a nuclear localization signal (Tag:NLS(Unk)).
Expression of SuUR495-962.Scer\UAS.T:Ivir\HA1,T:Uuuu\nls6 under the control of Scer\GAL4AB1 results in the formation of miniature salivary glands. The nuclei of the salivary glands cells are also significantly smaller than normal. Weak points in the polytene chromosomes are still seen in these animals.
Scer\GAL4arm.PS/SuUR495-962.UAS.Tag:HA,Tag:NLS(Unk) partially rescues SuURES
Expression of SuUR495-962.Scer\UAS.T:Ivir\HA1,T:Uuuu\nls6 under the control of Scer\GAL4arm.PS in a SuURES background results in approximately 1% of nuclei in larval salivary gland squashes having weak points in intercalary heterochromatin regions (this indicates some rescue of the SuURES phenotype of lack of underreplication).
