Expression of AhcyL1HM05009 driven by Scer\GAL4Act5C.Switch.PR (in males fed RU486 during adulthood, to avoid developmental effects) significantly extends lifespan compared to controls; at 40 days old, age-dependent decline in climbing ability (seen in controls) is significantly suppressed in RU486-fed males and age-dependent decreases in fecundity (seen in controls, measured as number of eggs laid per day) is significantly suppressed in RU486-fed females. There are no significant differences in survival of RU486-fed Scer\GAL4Act5C.Switch.PR>AhcyL1HM05009 flies with addition of paraquat to food, compared to controls (no increased ability to withstand oxidative stress). Expression of AhcyL1HM05009 driven simultaneously by Scer\GAL4bun-Switch1.32 and Scer\GAL4Switch1.106 (in males or females fed RU486 during adulthood) does not significantly affect life span compared to controls; when expression is driven by Scer\GAL4elav.Switch.PO or Scer\GAL4TIGS-2, there is a significant extension of lifespan in RU486-fed males and females. Dietary restriction (reduced sugar and yeast in food) significantly increases lifespan; in Scer\GAL4Act5C.Switch.PR>AhcyL1HM05009 flies, dietary restriction does not additionally extend lifespan.
