Expression of Dop1R2KK110947 driven in the dorsal fan-shaped body by Scer\GAL4GMR23E10 results in flies sleeping significantly more during the day and late hours of the night than parental controls; daytime sleep bouts are prolonged without affecting their frequency.
Focal dopamine application onto dorsal fan-shaped body neurons leads to hyperpolarization and suppresses spiking in controls; this suppression is significantly blocked in Scer\GAL4GMR23E10>Dop1R2KK110947 neurons. Prolonged dopamine application switches neurons in controls from electrical excitability (ON) to quiescence (OFF); this does not occur in Scer\GAL4GMR23E10>Dop1R2KK110947 neurons.
Expression of Dop1R2KK110947 under the control of Scer\GAL4dimm-929 does not affect sensitivity to (S,Z,Z)-CH503 (a more potent stereoisomer of the gustatory sex pheromone CH503). As in wild type flies, courtship defects are seen in response to (S,Z,Z)-CH503.
As in wild-type, female flies expressing DopR2KK110947 under the control of Scer\GAL4Gr5a.8.5 show an increased sugar-sensitive proboscis extension reflex in response to either L-Dopa feeding or 6 hours of wet starvation.
