The class IV dendritic arborization neurons in flies expressing Pka-C1Scer\UAS.cKa under the control of Scer\GAL4ppk.PG show a significant decrease in the complexity of dendritic arbors in respect to the total dendritic length. The total number of dendritic branching points is also decreased.

Flies expressing Pka-C1Scer\UAS.cKa under the control of Scer\GAL4fat have a reduced fat content compared to controls.

1.2% of egg chambers have at least one binucleate nurse cell in females expressing Pka-C1Scer\UAS.cKa under the control of Scer\GAL4nos.PG.

When Pka-C1Scer\UAS.cKa is driven by Scer\GAL4GMR.PF, the eye is discoloured and swelled up with wrinkles. The boundaries of all ommatidia and photoreceptor cells disappear.

Expression of Pka-C1Scer\UAS.cKa under the control of Scer\GAL430A results in a collapsed wing phenotype. The flies have melanotic growths in the head and show a developmental delay of about 2 days compared to controls.

When Pka-C1Scer\UAS.cKa is driven by Scer\GAL430A, wings are variably unfolded or unfolded and blistered, or collapsed. All patterns elements of the wing (veins bristles and hairs) are normal. Also melanotic tumours are often seen within the head capsule. Melanotic tumours are also found in the wing or other parts of the body. Occasionally dead or moribund flies are found that have become entirely melanized after partially opening their wings. When Pka-C1Scer\UAS.cKa is driven by Scer\GAL4684 or Scer\GAL471B, severe blistering of then wing blade is seen, accompanied by enlarged veins and a disrupted venation pattern.

Expression of insertion P{UAS-Pka-C1.K}15.3 in backgrounds Scer\GAL4RK5 and Scer\GAL4KO5 allows partial survival; 35 with a sibling class of 47, and 24 with a sibling class of 129 respectively. In the Scer\GAL4PP3 background 15.3 shows complete lethality. Expression of insertion P{UAS-Pka-C1.K}13.1 in the Scer\GAL4RK5 background allows partial survival; 3 with a sibling class of 46. In the Scer\GAL4PP3 and Scer\GAL4KO5 backgrounds 13.1 shows complete lethality. Animals weakly expressing Pka-C1Scer\UAS.cKa under the control of Scer\GAL4RK5 sometimes die as blackened pharate adults, but adults that eclose and have a variety of overlapping phenotypes are also produced. These phenotypes include warped or collapsed wings, flecks on the surface of the eye and bristles glued to the thorax. Flies that are as small as half the normal size are also seen. Some show a combination of these defects, others were normal. In the Scer\GAL4PP3 and Scer\GAL4KO5 backgrounds P{UAS-Pka-C1.K}16.5 show complete lethality. Animals weakly expressing Pka-C1Scer\UAS.cKa under the control of Scer\GAL4KO5 sometimes give rise to adults that eclose but have collapsed wings. Expression of insertions of P{UAS-Pka-C1.K} show complete lethality in the Scer\GAL4KO5, Scer\GAL4PP3, and Scer\GAL4RK5 backgrounds. Expression of insertions of P{UAS-Pka-C1.K} in a Scer\GAL4e22c background that show the strongest phenotype usually fail to hatch, and the majority develop normal larval denticle belts.

Expression of Pka-C1Scer\UAS.cKa under the control of Scer\GAL4fat suppresses the increased fat content seen in Lsd-11 flies.

The collapsed wing phenotype caused by expression of Pka-C1Scer\UAS.cKa under the control of Scer\GAL430A is suppressed by the presence of the transcriptionally active P-element in the P{}cactBQ insertion. The collapsed wing phenotype caused by expression of Pka-C1Scer\UAS.cKa under the control of Scer\GAL430A is suppressed by the presence of the P-Repressor encoded by P{P-Sal}89D. Mosaic analysis using the P-Repressor encoded by P{P-Sal}89D to repress expression of Pka-C1Scer\UAS.cKa under the control of Scer\GAL430A indicates that Pka-C1Scer\UAS.cKa must be expressed for the lethal interaction between bae1 and Pka-C1Scer\UAS.cKa expressed under the control of Scer\GAL430A to take place.

The addition of CrebB-17ACbz.Scer\UAS or CrebB-17AS162 has little effect on the Pka-C1Scer\UAS.cKa, Scer\GAL430A phenotypes. The addition of Stat92E06346 or Stat92Ej6C8 has little effect on the Pka-C1Scer\UAS.cKa, Scer\GAL430A phenotypes. The addition of Scer\GAL430A, strongly suppresses the wing phenotype caused by Pka-C1Scer\UAS.cKa, Scer\GAL430A alone. Fewer survivors are seen than expected, though - and those flies are weak and are trapped easily in medium. The addition of bae1 to Pka-C1Scer\UAS.cKa, Scer\GAL430A flies is lethal.

Comments

Expression of Pka-C1Scer\UAS.cKa under the control of either Scer\GAL4Mef2.247 or Scer\GAL4c309 almost completely rescues the temperature preference behavior defects seen in Pka-C1MB581/Pka-C1X4 mutant.
