Results from response to cervical stimulation, intracellular recordings of flight muscles and direct excitability of muscle demonstrate nerve conduction is intact but the evoked response gradually diminishes as the temperature is raised, there appears to be a defect at the neuromuscular junction.

Order of temperature sensitivity in the hemizygous condition: shi4 > shi21 > shi1 > shi2. Order of temperature sensitivity in the homozygous condition: shi21 = shi1 > shi4 >= shi2. Order of temperature sensitivity of heteroallelic combinations: shi2/shi21 = shi1/shi21 >= shi4/shi21 > shi1/shi2 = shi1/shi4 >> shi2/shi4.
