Abi has two ena binding motifs, each containing a critical LPPPP motif. Test of 95 amino acid changes to each LPPPP motif reveals that most mutations disrupt binding: first residue tolerates L or aromatic residues (FYW). Proline residues have little tolerance to change.

The Abi-ena interaction is required for lamellipodia formation in larval macrophages. Disruption of Abi binding to ena does not disrupt the Abi-ena interaction.
