Nucleotide change:

G634862A

Amino acid change:

W173term | Dredd-PD; W173term | Dredd-PE; W173term | Dredd-PF; W119term | Dredd-PG

Reported amino acid change:

W195term

Comment:

Nonsense mutation (TGG to TAG) at Trp codon.

Comment:

The predicted amino acid change was reported relative to a 516aa Dredd protein, which would be created by the use of an uptream translation start. The current Dredd genome annotation uses a downstream AUG which is supported by both species conservation and RNA-seq data. The two start sites are 22 aa apart.

Detailed Description

The programmed cell death of bursCCAP neurons seen in the adult ventral nerve cord of wild type flies after eclosion is strongly suppressed in DreddD55 mutant homozygotes.

DreddD55 homozygotes exhibit wild-type apoptosis of vCrz neurons, with no viable vCrz neurons detected at 7 hours after puparium formation.

Mutant flies that are infected with E. coli one day prior to infection with either X. nematophila or P. luminescens are highly susceptible to infection by either X. nematophila or P. luminescens (in contrast to wild-type flies which show increased survival after infection with either X. nematophila or P. luminescens due to partial protection by the prior infection with E. coli).

The proportion of DreddD55 homozygous pupae (24 hours APF) surviving to adulthood after UV irradiation (50,000 μJ/cm2) is significantly higher than for wild-type.

DreddD55/DreddD55 is a non-suppressor of eye phenotype of Scer\GAL4GMR.PF, egrUAS.cMa

Additional Comments
Genetic Interactions
Statement
Reference

DreddD55 PGRP-LBΔ double mutant adults infected with Erwinia carotovora carotovora by oral feeding exhibit an absence of immune response (immune deficiency pathway activation) in midgut and fat body compared to controls.

Homozygosity for DreddD55 does not supress the ablation of adult eye tissue seen in egrScer\UAS.cMa; Scer\GAL4GMR.PF animals.
