[32]P labeled pri-miRNA form of ban was processed to the hairpin pre-miRNA form using S2 nuclear extract and gel purified. This labeled pre-miRNA was used as a substate with Dcr-1 affinity purified under high salt conditions.

Dcr-1 cleaves ban pre-miRNA into the mature miRNA form. This is stimulated by purified GST-loqs protein and requires Mg[2+].
