ScerGAL4VGlut.VGN6341 drives expression mainly in the adult subsesophageal zone, with expression in ~12 neurons on each side of the adult pars intercerebralis that project into the subesophageal zone. In the adult subsesophageal zone there are two clusters of neurons each side of the midline, one medial and the other lateral. The medial cluster has 6 neurons on each side, 2 of which are motor neurons innervating muscles of the proboscis and the lateral cluster also has 6 neurons on each side, 3 of which are motor neurons innervating muscles of the proboscis, the remaining neurons of each are interneurons, including the bitter gustatory local interneuron. Expression is also seen in neurons, including leg motor neurons, of the thoracico-abdominal ganglion.
ScerGAL4VGlut.VGN6341 drives expression in 3-4 motor neurons that innervate the tibial depressor muscle and long tendon muscle 2 of the femur and 1-2 motor neurons that innervate the tarsal depressor muscle in the tibia.

Thermogenetic activation (using TrpA1UAS.(B).cKa at 31[o]C) of Scer\GAL4VGlut.VGN6341 neurons in adults results in a significantly reduced proboscis extension reflex in response to tarsal sucrose or water, compared to controls; the reduced response to sucrose is still seen when expression is restricted using Scer\GAL80tsh-GAL80, but not using Scer\GAL80ChAT.3.3P.
Inhibition (using shi1.UAS at 31[o]C) of Scer\GAL4VGlut.VGN6341 neurons in adults results in a significantly increased proboscis extension reflex in response to tarsal application of a sucrose and bitter mixture, compared to controls, with no effect on the response to sucrose alone.
